 On April 12, 2023, Dr. Xinyan Li was invited to attend The 22nd China Biological Products Annual Conference (CBioPC) and chaired the first half of the afternoon discussion of the Cell Therapy and Gene Therapy session on April 13. In this session, Dr. Xinyan Li gave a presentation titlled "Opportunities and Challenges in the development of AAV Gene Therapy Drugs Development".

On April 19, 2023, base on the official website of the Center for Drug Evaluation, NMPA (CDE), the Investigational New Drug (IND) application clinical trial application of AAV gene therapy drug of FT-004 from Frontera Therapeutics was accepted for the indication of hemophilia B (endogenous FIX activity ≤ 2%).

On April 26, 2023, Frontera Therapeutics announced that China CDE accepts Investigational New Drug (IND) application for another gene therapy product, FT-003, for the indication of neovascular age-related macular degeneration (nAMD).

As the company's first Fist-in-class gene therapy drug, FT-002 based on positive pharmacodynamic results and partial clinical safety data from from some IIT subjects, the pre-IND meeting application for FT-002 was submitted successfully on April 27, 2023. FT-002 indication is X-linked retinitis pigmentosa (XLRP).

Frontera Therapeutics Dosed First Patient for Gene Therapy drug candidate FT-003 for the Treatment of Diabetic Macular Edema (DME)

Frontera Therapeutics, a global, clinical-stage biotechnology company that seeks to develop novel and best-in-class gene therapy medicines to improve the lives of patients across multiple disease areas, today announced that it has dosed the first patient at the Ophthalmic Hospital of Tianjin Medical University, China, for its gene therapy program with its gene therapy, FT-003, for the treatment of Diabetic Macular Edema (DME).
DME is a severe retinal disease and is the main cause of vision loss for Diabetic Retinopathy (DR) patients. This represents a major breakthrough, as FT-003 is the first gene therapy product in China advancing into the clinical stage for this indication. FT-003 is, an AAV drug candidate developed independently by Frontera which delivers therapeutic anti-VEGF to the eyes, and is administered by a one-time intravitreal (IVT) injection.

CEO's Visit to US Site

From May 8 to May 20, our new CEO, Dr. Xinyan Li, flew to Boston, USA, to visit the Frontera USA team. Since the company was formded  three years ago, due to COVID-19, the teams in China and the U.S. could only communicate online. In spite of this, everyone overcame many difficulties such as the time difference to promote the project efficiently. It was the first time for Dr. Li to visit the US team, and she met with all the team members and had in-depth discussions on the work and projects.
Before returning home, Dr. Li attended a group building activity organized by the US team members at Revolution Hall, where the team shared the Chinese and American food, culture, and leisure lifestyle, and spent a wonderful afternoon together.
This trip to Boston officially kicked off the face-to-face visit between the US and Chinese teams, and also gave us a deeper understanding and knowledge of our US colleagues' work and lives, Looking forward to more visits between Chinese and American team!

What do we do in AD?
 First and foremost, analytical assay development. Our main responsibilities include (but are not limited to) the following
• Determine phase appropriate analytical method panel
• Develop critical analytical methods, qualify and transfer to QC, and support GMP activities
• Develop key analytical programs including RefStd, formulation, stability, specification, comparability, DP compatibility
• Establish relationship with CROs on programs at various stages
At the same time, we have lots of interaction with almost all the other teams and provide analytical support to the cross-functional program teams.

 In the past three years, we have successfully built the AD group and the capabilities of analytical development; including development, optimization and qualification of all analytical assays, supported research and process development, method transfer to QC, managed non-GMP stability studies, and supported IND submission for all the FT 1.0 programs. Our team has also played important role in the rhabdovirus free cell line development work.
Currently we are working on supporting all the FT2.0 programs, optimizing the AAV5 platform, developing new analytical assays for new programs as well as FT1.0 programs late stage development, supporting RD, PD, QC, Clinical, RA and move the programs forward. Look forward to another productive year at Frontera!

Our company has recently completed the first dosing of FT-003 for DME (Diabetic Macular Edema), which is certainly another major milestone for our company. But what is diabetic macular edema? To understand diabetic macular edema, we first need to understand DR(Diabetic Retinopathy), because DME can occur at different stages of DR. Today we will focus on diabetic retinopathy.
1. First, what is diabetic retinopathy?
Diabetic retinopathy is the most common retinal vasculopathy, one of the major blinding eye diseases in people over 40 years of age, and irreversible microangiopathy secondary to diabetes.
2. If you have diabetes, does it mean that there is no problem with your retina if you don’t feel vision decline?
 Here is an example: Mr. Li is a 40-year-old diabetic patient, diagnosed with type 2 diabetes. His BCVA of the right eye was always around 20/25, but recently his vision suddenly decreased and went to the doctor. Fundus photography showed that his right eye has already progressed to advanced diabetic retinopathy (proliferative diabetic retinopathy) with massive retinal hemorrhage, which needs to be treated with surgery.
Therefore, it is highly recommended that if you or a family member is diagnosed with diabetes, please go to see an ophthalmologist and check the retina regularly to detect the lesion and intervene early and prevent further vision loss.

3. What are the signs of diabetic retinopathy? Who with diabetes can be diagnosed with diabetic retinopathy?
  Diabetes brings about lesions in blood vessels throughout the body, and delicate organs like our eyes cannot escape. There may be loss of vision, distortion of vision, black shadows in front of the eyes, and in severe cases, loss of vision to no light perception. Some diabetic patients feel that they have good vision, but the actual lesions in the retina may have already reached an advanced stage, with a large numberamount of neovascularization or fibroproliferative traction, but the retinal structures in the macula still have normal functions. However, in this case, once the neovascularization occurs, secondary retinal detachment or massive hemorrhage will occur, and vision will be suddenly lost.
4. Prevention and treatment of diabetic retinopathy
    First of all, control of blood glucose, and blood pressure, and timely treatment of eye lesions have an important impact on the prevention and development of diabetic retinopathy. Diabetic retinopathy can develop gradually from non-proliferative to proliferative, and once the transition to proliferative diabetic retinopathy occurs, vitreous hemorrhage and retinal detachment can occur at any time, which can eventually lead to blindness. Macular edema is another common manifestation of diabetic retinopathy, which is one of the important factors leading to severe visual impairment in diabetic patients.
     Secondly, treatments for DR include retinal laser photocoagulation and anti-VEGF therapy, and so on. In cases of recurrent vitreous hemorrhage and retinal detachment, vitreoretinal surgery is usually required.
      So as soon as you are diagnosed with diabetes, please get an ophthalmic examination to check your retina and intervene in DR if needed.

Looking back on this project, two things impressed us most: COVID-19 pandemic caused time-outs in the review of pre-IND and the submission form changed from paper to electronic. 
December 2022 is the most critical time for FT-003 filing while COVID-19 is spreading like wild fire across China. According to our timeline, we would receive CDE  comments for FT-003 pre-IND on December 12, 2022, and after that, we need to respond to CDE comments one by one and revise IND dossier accordingly. We have to submit IND application prior to  January 9, 2023 or the start of phase I clinical trial may be affected. However, almost all CDE staffs were infected by COVID-19 at that time, so FT-003 pre-IND was not reviewed and replied. After dialing CDE’s telephone several times, we found that their telephone was disconnected, we had to give up the call, fell into endless waiting and anxiety, and seeking solutions in WeChat communication groups.

On 26 April 2023, FT-003 Injection obtained the notification of clinical trial.。
Despite a series of challenges during FT-003 IND kick off in March 2022 and IND approval in April 2023, such as Shanghai lockdown, COVID-19 pandemic all over the nation, staff adjustment of the company, and the change of the submission form, we still obtained the notification of clinical trial more than a week earlier than expected (the following figure is the timeline we set in July 2022). 

In addition to the delay of pre-IND response, which would result in the inability to finalize the IND dossier, there is another difficulty: Beijing Express is almost paralyzed due to the COVID-19 pandemic, so our document cannot be successfully mailed to CDE. FT-003 is one of the most valueable projects in our company, and the review progress affects everyone on the team. At the weekly Core meeting team, the U.S. team also helped us finding solutions. Ting Yang (Project Leader) even advised us sending the dossier to reviewer’s home by ourselves. Although this suggestion was not taken, we were moved by everyone's enthusiasm. 
Finally, on January 4, 2023, we received pre-IND responses from CDE, more than 20 days later than we expected!!! At this timepoint, we only have five days left from the scheduled submission date...... what is more challenging is that almost everyone involved in this project has not recovered from COVID-19. The very next day, we pulled the project members to meet, discuss, draft responses, revise IND dossier over and over again,. Finally, it took us only 4 days to finalize all the dossier. 
The last issue before IND filing is the change of submission form. Previously, paper submissions were required, but now the submission form changed into electronic submissions. Although we no longer need to print and bound paper document, there is also a lot of new requirements for electronic submissions, such as "electronic signature" and" signature verification ", which we do for the first time. And since these are new requirements, there is no clear guidelines we can refer to, we have to fumble and operate, trembling to complete the above work, and sent out this valuable CD-ROM documenting the efforts of all the teams with an uneasy heart.
After 60 working days of review by CDE experts, FT-003 Injection IND submission was finally approved. This exciting milestone is mainly due to close cooperations of each department of the company, demonstrating the strong cohesion and excellent executive capability of our team.

In order to enable DP Prodcution capabilities and Standardize DP opetations,  Suping Zhao from the Production Department DP Section recently conducted a training for colleagues in the production department who will perform the filling operations in the future, which is briefly summarized as the "three key techniques" of formulations.
1. High awareness
Operations inside the isolator chamber have big impact on the sterility, foreign objects, and other quality aspects of the product. Therefore, it is not allowed to act upstream of the airflow at the opening of the drug solution and packaging materials. The action should be gentle to avoid interfering with the airflow pattern, and unnecessary actions should be strictly avoided.
2. Accurate operation
Sterile operations should be strictly carried out in accordance with the validated actions, including methods and frequency, to avoid any errors caused by human factors.
3. Maintain a positive attitude
It is necessary to continuously strengthen the practice of operational techniques, improve aseptic awareness, precision awareness, enhance business capabilities, strictly inspect and audit, and strive to achieve zero errors and zero errors.
Training is only a short-term professional training, but we also need to improve our regular learning mechanism and form a long-term self-learning and self-checking work and learning style. Training on professional knowledge should follow the principles of "authenticity, accuracy, and systematicity" to clarify the scope, techniques, and steps of equipment operation. Only then can we strive to be the leader and vanguard in work and learning.

In order to highlight the importance of standardized handwashing in clean areas, the trainer made comparison samples and formed pictures with professional colony counters, objectively and vividly demonstrating the "7-step handwashing method" and the significance of the correct use of disinfectants.
In order to let everyone more vividly understand different types of bacteria such as " coccus, bacilli, mold", the trainer used dyeing identification technology and combined with digital microscope to turn these different types of bacteria into pictures, so that everyone can identify them more intuitively.
To underscore the importance of disciplined behavior in clean areas, the trainer used objective data to illustrate the big impact of some common small actions, Any inconspicuous action can damage the environment of the clean area, remember "Don't assume that things can be done with small disadvantages";

The evaluation and approval of contracted analysis service supplier is an important part of GxP management, which impact data reliability of registration dossier directly. 16 on-site or remote audits of 11 suppliers have been completed by QA department. By comprehensively evaluation of organizational structure, experimental conditions, instrument management, personnel qualification, training, reagent, reference standard management, analytical records, etc. Total 16 audit reports were completed including nearly one hundred observations. The corrective and preventive actions of the observations will be tracked by CAPA management system. Ensure that all the contracted testing activities comply with GxP regulation.

In the biotech industry, cell culture is an important step in manufacturing. In the cell culture process, the shaker is an indispensable and irreplaceable instrument which is of ingenious design, exquisite technology, precise control, and the most important expensive.
One day, the engineering equipment department received a work order from the user to repair a certain brand of shaker, and the description was that the shaker door did not work properly. EE engineer got the notice and did an onsite inspection immediately. At the same time, the engineer also contacted the vendor for more professional support.

During new instrument installation, we established a good relationship with the shaker vendor. We also solved the shaker bug for their Beijing headquarter. So, when they got informed, they responded immediately. They dispatched an engineer to our Suzhou site for an on-site inspection. The EE engineer and the vendor engineer had a discussion. Both agreed that the malfunction could be caused by the screws loose and mechanical locker malposition. EE engineer tightened the screw, the malfunction got fixed.

so he disassembled the shaker shell, and after many tests to open and close the door, carefully observed the running path and method, and found that when the door was opened to a certain angle, the air brace instantly produced a very slight deformation, changed the running path, and left a scratch near the left door air brace. Compared with the right door air strut of the same structure, there is no such scratch. The engineer suspected that the cause of the failure was insufficient support for the air strut to prevent the door from opening and closing properly. During this period, the engineers visually found that the center position of the entire structure had a slight offset in the process of opening and closing the door mechanical lock. Compared with the right side, the mechanical lock structure of the entire door is strong and stable, and there is no deformation and flutter during the process of opening and closing the door. Therefore, this doubt was communicated and demonstrated with the brand engineer and the user, and the user felt that the two sides were running differently, which may be the cause of the failure. At this time, the brand engineer removed the air strut for inspection according to the maintenance experience, and found that the 150N air brace engineer could easily press down. The engineer confirmed that the cause of the fault was that the air strut support force was insufficient, resulting in the inability to open and close the door normally, and arranged for the Beijing headquarters to express the air brace spare parts to Suzhou.

After investigations and comparisons, combined with the actual conditions, the emergency escape automatic door was selected. The original function was improved, and the door has following advantages:
1. In the case of closing the door, with locking function, it cannot be manually opened from the outside to meet the anti-theft function.
2. Under normal circumstances, it is used like an ordinary automatic swing door to meet the passage function.
3. In case of emergency or power failure, the door leaf can be completely pushed from the inside to the outside to the maximum opening, the door page can be opened directly vertically 90 degrees, and the movable fan and the fixed fan can coincide at the same time, and the opening amplitude is doubled, which is conducive to the emergency evacuation of the crowd and convenient for escape.
4. When transporting bulky items, you can manually push the fixed fan outward, and the opening range is doubled, which is convenient for transporting bulky items.
Auto-doors provide easy access and also some kindly remind as below:
1. When the automatic door is operating normally, please do not stop when the automatic door is open, so as not to be injured by the automatic door.
2. When the automatic door is closed, do not use your hands to forcibly open and force through, so as not to be injured by the automatic door.
3. Do not stand inside the door frame and chat to avoid being caught by the automatic door.

The door at reception hall is the first guardian of the company's security. It ensure security for whole company, but there were some issue for the existing door. Wind could be easily bending the door leaf, away form its original location. There was also a gap between the jamb and the framing, so air conditioning and heating seem not working in the hallway. Insects and Reptilia could enter the facility from under the door. The main door displaced a lot of functions. It has been upgraded during the Labor Day holidays.

When we are approaching the completion of rBV generation for pipeline programs FT-011, -017 and -018, RD team would like to give a big shoutout to our junior scientists on our team, Matt, Amy and Brendan. Matt’s contribution is especially valuable for this achievement, his pioneer work paved the road for us to generate research grade rBVs in scale for all the programs besides AAV capsid improvement and optimization. While working on more challenging constructs of FT-011, Amy not only demonstrated her persistence, but also her excellence in documentation, which is particularly important when we are generating materials for DC nomination. Brendan brought much expertise from AD group to us, he is not only talented in running complicated assays, but also establishing new assays efficiently, and led to our many process evaluations completed with quick turnaround time. The efficient and smooth teamwork of Matt, Brendan and Amy has now become the most important force in RD team to provide quality materials for our pipeline programs. We look forward to seeing their work contributing to the success of AAV production and in vivo studies!

Interview with Linrui Li from Analytical Development, who was nominated for Star Award recently:
1. Please tell me something about yourself.
I joined AD in 2021 after graduating from Northeastern University. This is my first job and I joined when Covid Pandemic was at its peak. The company was still in Newton, the old location.
2. You won the Star Award for your extensive involvement in HPLC, NGS, CE, ELISA and ddPCR work. How do you feel after you receive the award?
I joined the company with a background in Bioinformatics. To learn and master these skills takes time and effort. I’m glad I was able to do it. I like fun and challenging stuff.
3. What do you do outside work?
I like traveling, to see different cities and learn different cultures. I have been to cities like New York, Washington DC, Orlando, Seattle, San Francisco, Los Angeles and cities in Alaska.

Star Award Interview

The hectic pace of life may involve stopping for food, or tasting it, or simply enjoying the process of making it. Today I would like to share with you a dish for all ages: spicy abalone chicken.
INGREDIENTS AND SEASONINGS
Ingredients: 500g chicken drumstick, 12 small abalones, Favorite vegetables (suggested to choose the vegetables is not easy to overcooked, I generally put green beans).
Seasoning: garlic clove, millet pepper, spicy soybean paste, oil consumption, soy sauce, white granulated sugar.
PROCEDURES:
1. Chop the chicken drumstick, wash the abalone that removed shell and gut, peel the garlic cloves, dice the millet pepper, Cut the green beans into sections.
2. Pour 2 spoonful of spicy soy sauce, 1 spoonful of oil, 1 spoonful of dark soy sauce, 1 spoonful of sugar in a small bowl, stir well and set aside.
3. Put the drumsticks in the pot before the water comes to a boil. Skim off the foam when the water comes to a boil.
4. Pour oil Into the pot. When the oil turn to smoke, add garlic stir fry fragrance, put the drumstick block and abalone into the pot. When the drumstick block stir fry to the surface slightly golden, add prepared sauce stir fully, add spicy millet, add the right amount of boiling water (water does not exceed the amount of meat, it is best to join after leaving a peak), Put the green beans into the pot, turn medium fire stew for 30min. After 30min later, add other additional seasonings according to your own taste.
5. Finally turn the heat to collect the juice, and we're done.

Tips to Improve Efficiency

Do you often fall into the trouble in the picture above? You are expected to complete one task and then the next with ease, but in reality you are often forced to deal with multiple tasks at the same time, and you are suddenly attacked by the ancient tasks left behind by the very procrastination (who is monitoring me?)

How can I get rid of this passive state? How can I handle multiple tasks with ease? How can we say goodbye to "procrastination"?
Here are a few tips to help you improve your work efficiency.

Fourth, refuse to procrastinate:
 Many times procrastination is due to the fear that the work will need to be "reworked", so the perfectionists always polish long time to delay until the task deadline to deliver, but the demand side will still ask for "rework", and eventually become that "from the A long-ago task that sneaks up on you from behind"
1. Create a sense of "minimum deliverable" and produce the most rudimentary version that you can show to the demand side early on;
2. Seek feedback from the other side, then iterate it to ensure that there is always only one task in hand (more iterations of important things, first iterations of urgent things)

For long vacation such as Labor Festival and National Festival are popular times for cycling around Taihu Lake, with a cycling mileage of about 270KM and a cycling time of about 14 hours. It's a great challenge for non-professional cyclists. For me, it is not only a physical self-challenge, but also a self-challenge in perseverance.
Road bikes and mountain bikes are recommended as riding tools, of course, as long as you are brave enough to ride a car, you can choose from the bike, but the two-legged 11-way is not recommended! 
When you have a car, you need to be mobile, bring a cycling helmet, gloves, cycling clothes, sun protection, tire repair and full set of tools. Energy supplies, mobile phones and power banks are also essential, and you are ready to go.
Cycling Taihu Lake is a long-distance ride, if you want to ride, you must do your physical reserves and do what you can. A budget with good fitness can be completed in one day, and if you are not fit, then prepare to complete the lake in two days. If you are not in a hurry, it is recommended to complete it in three days, and you can enjoy the beautiful scenery along the way.
Meet cycling enthusiasts on April 29, depart from home at 4:30 a.m., bring energy drinks, chocolate bars, and ride all the way under the guidance of street lights. First ride 60KM to reach the first resting point of Wujiang at about 7:00, which is also the place to eat breakfast, a bowl of stewed meat noodles. Take a short rest, continue to set off, basically rest every 30KM for the whole journey, and arrive home at 7 pm. Plan your rest points and strategies before you go, so you won't be embarrassed during the journey around the lake.
