The Protein Science and Production Week
In January, our US team exhibited at PepTalk 2024 in San Diego. We had the privilege of engaging with numerous attendees, and we extended our warm congratulations to the winners who walked away with their llama prizes. Peter Hsueh, PhD., who presented an insightful poster titled 'Product and Impurity QC for Reagents Used in Biopharma Manufacturing' (Poster B02) was the highlight of the event. His valuable knowledge and insight shed light on the crucial quality control aspects of reagents utilized in biopharmaceutical manufacturing. We are proud of our team's accomplishments and the positive engagement they received at the event.

In December 2023, Beisen announced its annual list of Best Employers for 2023, and ACROBiosystems received the 2023 Best Employer Management Practices Award.
The award recognizes the milestones that ACROBiosystems has been achieving in its continuous efforts and innovative practices to enhance the impact of the employer brand. ACROBiosystems is dedicated to accelerating the drug development process and helping to bring them to market. Our mission is to actualize the clinical application of novel, innovative therapies and contribute to the biomedicine industry. Through our efforts, we would like to start by bringing value to our customers to creating value for the whole industry in the global market. The company continues to actively empower its partners by updating its technology, products, and services. In this way, we can accelerate the R&D and marketing process of various biopharmaceuticals, including target drugs and immunotherapeutic drugs, as well as better clinical applications, and ultimately contribute to the cause of human health.

From being a newcomer to a valued team member, I could sense my progress and integration within the team. Collaborating with colleagues, we completed various projects, contributing to the company's development.
The New Year signifies new beginnings and possibilities. At ACRO, I realize that each day presents a fresh opportunity for innovation and progress. I encourage myself to remain curious, bravely try new methods and perspectives, and bring creativity and vitality to the team.

Introduction
At present, viral vectors are mainly developed from four types of viruses: adenovirus, adeno-associated virus (AAV), lentivirus, and retrovirus. Adenoviruses and AAV efficiently infect host cells in free form and express large amounts of the proteins encoded by the foreign genes they carry. In contrast, retroviruses and lentiviruses integrate into the host cell genome. These vectors do not kill host cells and can be used to stably transfect mammalian cells. No single cell line can be infected by all viral vectors, and retroviruses have strong species specificity and a clear preference for dividing cells.
What is a lentiviral vector?
Lentivirus vector (LV) is a kind of vector derived from retrovirus. The reason why lentivirus is called lentivirus is that the main clinical characteristics of infection are a long incubation period before the appearance of typical clinical symptoms and then slow onset. The most classical lentivirus is modified from HIV, and the HIV-1/HIV-2 system has also been widely used. Subsequently, there are simian immunodeficiency virus (SIV) vector systems, felines immunodeficiency virus (FIV) vector systems, and so on.
The development of lentiviral vectors
Lentivirus is a gene therapy vector based on HV-1. The first-generation two-plasmid system lentiviral vector encodes all packaging elements in a single vector with high biosafety risk. The second generation of three-plasmid lentiviral vectors deleted all HIV accessory genes vif, vpu, vpr and nef to reduce the risk of RCR production. Replication of the promoter activity did not affect virion titer. The 3’LTR of the U3 region was deleted in the third generation of the four-plasmid system lentivirus vector so that the vector lost the HIV-1 enhancer and promoter sequence and could not transcribe RNA even in the presence of all viral proteins. At the same time, the tat gene was removed and replaced by a heterologous promoter sequence. The third generation is the currently popular lentiviral packaging vector.

Advantages of lentiviral vectors
√Once a lentiviral vector infects a cell, the RNA genome of the virus is reverse transcribed into DNA, which is then permanently integrated into the host genome, thereby allowing long-term stable expression of the genes carried on the viral vector. That is to construct stable cell lines.
√Lentiviral vectors are capable of infecting both dividing and non-dividing cells.
√Lentiviral vectors could infect many types of cells, such as neuronal cells, hepatocytes, cardiomyocytes, tumor cells, endothelial cells, and stem cells.
Packaging of lentiviral vectors
To package a complete lentivirus, we generally need the following plasmids:
① A shuttle plasmid: this is the "protagonist" carrying our target gene in the LTR region of both ends, but it is replication-defective, so it is not enough alone, and the assistance of the following plasmid is needed.
② One or two packaging plasmids
③ An envelope plasmid
Then these plasmids were co-transferred into 293T cells, and the packaged virus would be released into the supernatant. The viral supernatant was collected and concentrated (or you cannot concentrate, directly use the supernatant), and then we could infect our target cells.

Application of lentiviral vectors
√Protein expression, stable strain construction. With the help of the ability of lentivirus to integrate sequences into the host genome to complete stable gene expression.
√CAR-T, CAR-NK, and other cell therapy. The use of viral vectors to transfer specific CAR genes to T cells or NK cells, lentiviral vectors are the current popular choice.
√Lentivirus vector-mediated RNAi technology. LV-RNAi mediates RNAi transfection efficiency and expands the range of vector-infected cells, which can be used to study cell-specific gene function and gene therapy.
√Gene editing. Genetic modification including gene therapy, CRISPR, etc., can involve lentiviral vectors.
√For the construction of pseudo viruses. It is applied to the detection of serum-neutralizing antibodies and lays the foundation for the subsequent screening and evaluation of monoclonal antibodies and antiviral drugs, vaccine development, and virus infection mechanism research.

Lentiviral vector infects cells
After packaging, the lentivirus can be used to infect cells. First, lentivirus enters cells through membrane fusion or endocytosis to release RNA. Then, through the action of reverse transcriptase and integrase, lentivirus forms a DNA reintegration complex to enter the nucleus and randomly integrate into the genome of the host cell and then starts transcription and translation.

It would be really helpful to indicate the storage requirements on the outside of the box. We had a close call where the package was almost left out overnight simply because we didnt know exactly what was inside and HOW it needed to be stored upon receipt. I highly encourage you to consider this. If we are going to keep ordering from you all, this is definitely something we would expect moving forward especially because most vendor we deal with, do this.
Do you have any suggestions and feedback?
We really like to products from Aero, they deliver timely and good quality.
Very quick to respond to a request for a quote or questions. But a bit overwhelming when they insist multiple time to ask about the becoming of the quote. Tho still very friendly.
The most recent order was from a new client who could not find his antigen at a reasonable price through other vendors. I have purchased reagents from ACRO during the past 8 years and the products are always very high quality.
